Abstract Objective To establish a method of inducing proteinase production of Penicillium marneffei in vitro and to measure its proteinase activity. Methods The B-6323 strain of P. marneffei was cultured in proteinase inducing medium under different pH conditions, and the production of proteinase over the course of several days under different pH conditions was measured using a spectrophotometric method ( 280 nm UV light) . Results Proteinase was induced and it’s activity was measured over the course of several days under different pH conditions. The strain had the highest proteinase activity on day 6 at pH 4. 0; the highest proteinase activity on day 3 at pH5. 6; and no proteinase activity at pH7. 2 on any day. Conclusion A method of inducing proteinase production of P. marneffei in vitro and measuring its activity was established, and may be amethod to be used for future research to understand the biochemical properties of proteinase.
